Physiological Functions of Bacterial “Multidrug” Efflux Pumps

چکیده
Bacterial multidrug efflux pumps have come to prominence in human and veterinary pathogenesis because they help bacteria protect themselves against the antimicrobials used overcome their infections. However, it is increasingly realized that many, probably most, such physiological roles are distinct from protection of administered by humans. Here we undertake a broad survey proteins involved, allied detailed examples evolution, energetics, structures, chemical recognition, molecular mechanisms, together with experimental strategies enable rapid economical progress understanding true roles. Once these established, knowledge can be harnessed design more effective drugs, improve existing microbial production drugs for clinical practice feedstocks commercial exploitation, even develop sustainable biological processes avoid, example, utilization petroleum.

Biochemistry of Bacterial Multidrug Efflux Pumps
Bacterial pathogens that are multi-drug resistant compromise the effectiveness of treatment when they are the causative agents of infectious disease. These multi-drug resistance mechanisms allow bacteria to survive in the presence of clinically useful antimicrobial agents, thus reducing the efficacy of chemotherapy towards infectious disease. Importantly, active multi-drug efflux is a major mec...
متن کاملCurrent Advances in Developing Inhibitors of Bacterial Multidrug Efflux Pumps
Antimicrobial resistance represents a significant challenge to future healthcare provision. An acronym ESKAPEE has been derived from the names of the organisms recognised as the major threats although there are a number of other organisms, notably Neisseria gonorrhoeae, that have become equally challenging to treat in the clinic. These pathogens are characterised by the ability to rapidly devel...
متن کاملInhibitors of bacterial multidrug efflux pumps potentiate antimicrobial photoinactivation.
Antimicrobial photodynamic inactivation (APDI) combines a nontoxic photoactivatable dye or photosensitizer (PS) with harmless visible light to generate singlet oxygen and reactive oxygen species that kill microbial cells. Cationic phenothiazinium dyes, such as toluidine blue O (TBO), are the only PS used clinically for APDI, and we recently reported that this class of PS are substrates of multi...
متن کاملBacterial Multidrug Efflux Pumps: Much More Than Antibiotic Resistance Determinants
Bacterial multidrug efflux pumps are antibiotic resistance determinants present in all microorganisms. With few exceptions, they are chromosomally encoded and present a conserved organization both at the genetic and at the protein levels. In addition, most, if not all, strains of a given bacterial species present the same chromosomally-encoded efflux pumps. Altogether this indicates that multid...
متن کاملHow to Measure Export via Bacterial Multidrug Resistance Efflux Pumps
Bacterial multidrug resistance (MDR) efflux pumps are an important mechanism of antibiotic resistance and are required for many pathogens to cause infection. They are also being harnessed to improve microbial biotechnological processes, including biofuel production. Therefore, scientists of many specialties must be able to accurately measure efflux activity. However, myriad methodologies have b...
